Title: Research of a Rapid Quantitative Detection Technology for COVID-19 Neutralizing Antibodies Based on Side Flow Immunochromatography and SmartphoneAuthor: LIAN YE1, LUO LE YANG1, LIN QIAN CHENG1, ZHENG RUI YING2, ZHANG NA2. 1. School of Laboratory Medicine (School of Life Sciences), Wenzhou Medical University.[Abstract] Objective: This study aims to develop a rapid on-site quantitative detection technology for COVID-19 neutralizing antibodies (COVID-19 NAb) based on lateral flow immunoassay and smartphones. Method: By preparing the key raw materials RBD and ACE2 protein for COVID-19 NAb testing, a rapid on-site test card for COVID-19 NAb side flow immunochromatography was prepared, and a high-throughput COVID-19 NAb latex immunoturbidimetric method was established and evaluated for use in a fully automated biochemical analyzer. Based on this, an application called COVID-19 NAb Test App was developed. By using a portable reader to take T-line photos of the test card and analyzing the non-linear fitting algorithm of Spline, rapid on-site quantitative detection of COVID-19 neutralizing antibodies was achieved. Result: The intra batch CV value of this method is 0.88% to 9.70%, indicating good precision; The lowest detection limit (LOD) is 35 ng/mL and the lowest quantification limit (LOQ) is 75 ng/mL, indicating high sensitivity; The linear range is 75-40000 ng/mL, and the linear regression equation is y=0.9979x-86, R=0.9995; Compared with the latex immunoturbidimetric method, the average value measured by the latex immunoturbidimetric method (y) is 5692 ng/mL, and the average value measured by this method (x) is 5591 ng/mL. The regression equation is y=1.0411x-123, R=0.9982,After paired t-test, there was no statistically significant difference between the two methods (P>0.05). Conclusion: The research results indicate that the COVID-19 NAb Test App developed by the project has good precision, sensitivity, and linear range, and has consistent results with latex immunoturbidimetry. The research of the project provides a feasible solution for on-site rapid quantitative detection of COVID-19 NAb, and has the potential for on-site rapid quantitative detection of other biomarkers.[Keywords] COVID-19, Neutralizing antibodies, side flow immunoassay, smartphone, rapid quantitative detection.
